  1. Vedolizumab

Vedolizumab is a monoclonal antibody being developed by Millennium Pharmaceuticals, Inc. for the treatment of ulcerative colitis and Crohn's disease. It binds to integrin α4β7. The cell line was discovered by Dr. Andrew Lazarovits and Dr. Robert Colvin as the murine MLN0002 homologue. Their discovery of the mouse equivalent of this antibody—originally applied to anti-rejection strategies in kidney transplantation—was published in the journal Nature, in 1996. The research was conducted at Massachusetts General Hospital in Boston, MA. Dr. Lazarovits and Dr. Colvin's cell line was subsequently licensed to Millennium Pharmaceuticals of Boston for further development. Vedolizumab has been in Phase III clinical trials since 2009, and has recently met critical endpoints. Reports indicate that Vedolizumab was found safe and highly effective for inducing and maintaining clinical remission in patients with moderate to severe ulcerative colitis. It looks like it will be an effective abiologic agent without some of the toxicity issues previously seen with anti-TNF drugs. Vedolizumab is often associated with the α4-integrin drug, Tysabri. They are, however, entirely different drugs, with separate mechanisms of action. Although there are symptoms of progressive multifocal leukoencephalopathy found in Tysabri, Dr. Brian Faegan, head researcher, reported an absence of any instances of PML in the Gemini Trial, which is a particularly important finding.
